COMPONENTS
Each white, 40 x 30mm cylindrical polyurethane sponge contains 20 mg chronolone (flugeston acetate).
PHARMACOLOGICAL PROPERTIES
Chronolone is a synthetic steroid with a strong progestagenic effect similar to corpus luteum-derived progesterone.
The chronolon contained in the CHRONOGEST CR sponge is absorbed through the vaginal mucosa and reduces the secretion of GnRH from the hypothalamus and gonadotropins from the pituitary. Thence, maturation of follicles in the ovaries and ovulation are prevented. The drug shows its effect by binding to steroid receptors in target tissues, as with other steroid hormones. The sponge, which slowly releases chronolone, suppresses anger as long as it remains in the vagina. After the sponges are removed, gonadotropin hormone (PMSG) is applied to stimulate the spontaneous onset of the follicular phase in the animals. Thus, estrus is synchronized and ovulation, fertility and fertility are increased. Chronolone is easily absorbed during the 12-14 day intravaginal administration period. While Tmax varies between 6-22 hours, Cmax is between 1.11 and 2.27 ng/ml. It reaches a constant level in a short time following the start of treatment. Plasma chronolone concentration is constant for 9 days and then begins to decline. After removal of CHRONOGEST CR, plasma chronolone concentration is below 0.78 pg/ml (LOD). Plasma chronolone concentration throughout administration is sufficient to prevent ovulation and induce estrous synchronization after sponge removal. Afterwards, its plasma concentration decreases rapidly and chronolone is metabolized into hydroxylated metabolites.
AREA OF USE/INDICATIONS
It is used for estrus synchronization and ovulation induction in sheep and goats.
USAGE AND DOSAGE
In sheep and goats, CHRONOGEST CR is applied intra-vaginally with the help of an applicator. Dosage; One sponge per animal, regardless of body weight, breed, type and season. The application period is 12-14 days for sheep and 11 days for goats. At the end of the application period, the rope of the CHRONOGEST CR controlled release vehicle is removed by slowly pulling.
CHRONOGEST CR usage method:
A single sponge is placed into the vagina with a special applicator. The applicator should be disinfected with a suitable disinfectant before each application. The sponge remains in the vagina for 12-14 days in sheep and 11 days in goats. For synchronization and induction of ovulation, intramuscular injection of PMSG mixture α is recommended in sheep immediately after sponge removal. In goats, synchronization is induced with an injection of PMSG+Prostaglandin F2ₐ 48 hours before sponge removal. In sheep and goats, ovulation occurs 36-72 hours after the sponge is removed. PMSG dose varies between 400-700 iU depending on age, race, season, etc.
Placement of Sponge
- The applicator is disinfected with CHRONOGEST CR/Plastisept or a similar disinfectant containing quaternary ammonium salt before each application.
- The sponge is squeezed and placed into the obliquely cut mouth of the tube, with the knot point of the thread in front.
- During placement the animal is kept calm and motionless. The thread is taken out of the applicator and the pusher rod is inserted into the tube.
- The applicator is carefully placed in order to reach the cervix.
- Keeping the pusher rod stationary, the tube is withdrawn 2-3 cm and the sponge is allowed to come out of the tube. The pusher rod and tube are removed, leaving the thread of the sponge hanging down from the vagina to facilitate retrieval.

EXCESS(Ram Participation)
Insemination is done at 36 and 72 hours (55 hours in artificial insemination) after removal of the sponge. The method that gives the best results is the “adjective in hand” method in which the ewes(sheep) are shown to the ram one by one. Outside the season of breeding, 1 ram is used for every 5 ewes. During the season of breeding, 1 ram is sufficient for 10 sheep. Both adult and immature young ewes should not be shown to the same ram on the same day. Rams should rest for 10 minutes between 2 overs. Rams of some breeds may not provide service outside of the mating season. In this case, the sexual activities of these rams can be stimulated by showing them to 1-2 ewes in rutting 10 days before the sponge application. Sheep are brought into rutting with the application of 5 mg subcutaneous estradiol. Ewes in rut are shown to the ram twice a day, in the morning and in the evening (for 1-2 days), and only 1 mating is allowed. If there are many sheep groups to be treated, a program is made to work with the group every 8-10 days (every 3-4 days during the ram breeding season).
SPECIAL CLINICAL CONDITIONS AND SPECIAL WARNINGS FOR TARGET SPECIES
It is not for the treatment of infertility, it is only used in reproductively capable sheep and goats. Animals to be administered progestagen should not have genital and urinary tract infections. PMSG should not be applied to sheep before sponges are removed. Sheep and goats should be free from unnecessary stress such as sudden feed changes, vaccination and branding before and after insemination. If the thread is not visible when removing the sponges, the inside of the vagina should be investigated. An unpleasant-smelling liquid may be found on the removed sponge. This is a buildup of vaginal mucus and has nothing to do with the health or fertility of the animal. CHRONOGEST CR/sponge is applied only with its special applicator. The applicator is wiped and disinfected before each application. Alcohol, cresol, phenol and iodine disinfectants are not used.
Females:
- Sheep are prepared by flushing method (starts 1 week before sponge placement, continues until 3 weeks after mating)
- Vaccination and parasite control should be completed 1 week before sponge application at the latest.
- Ewes should be accustomed to the presence of rams. Especially if the sheep and rams are of different breeds, the acclimation process is useful to prevent problems during breeding.
- Since animals that come into heat again (return) can occur at any time, a ram joins the herd 15 days after the end of the process.
- Stress, sudden changes in environment and feed, antiparasitic treatment, vaccination, etc., which cause embryonic deaths and reduce fertility and birth rates,these situations should be avoided for 4 weeks after the mating of the ram.
- If artificial insemination is to be performed, it should be done 55 hours after the sponge is removed.
- There should be at least 75 days between the ewe’s last birth and sponge placement.
- There should be at least 150 days between the goat’s last birth and sponge placement.
- CHRONOGEST CR should be administered to ewes with vaginal discharge and recent abortions only after appropriate treatment.
Males:
- Rams with proven fertility should be used.
- Starting 6-8 weeks before mating, a balanced ration should be applied and stress factors should be avoided.
- Sponge placement should be scheduled to match the condition of the rams.
UNDESIRED EFFECTS
As a result of continuous progestagen applications, cystic endometrial hyperplasia and pyometra may occur in sheep. Rarely, vaginitis due to adhesions between the sponge and mucous membrane may occur in sheep and goats where the application is applied.
DRUG INTERACTIONS
Other reproductive drugs should not be applied during the application of sponges.
SYMPTOMS, PRECAUTIONS AND ANTIDOTE IN OVERDOSE
Although overdose is not possible due to the method of administration, leaving the sponges in the vagina for longer than the recommended time may cause local tissue problems.
WARNINGS ABOUT DRUG RESIDUES IN FOOD
Drug Residue Withdrawal Period (D.R.W.P):
Animals should not be sent for slaughter as long as the sponge remains in the vagina and for 5 days after withdrawal. The residue purification period for milk is zero days.
CONTRAINDICATIONS
Use during pregnancy: It is contraindicated as it may cause abortion in pregnant animals. For sheep that give birth before 75 days and for goats before 150 days, for those that abort immediately after,
It is contraindicated to apply to sheep with vaginal discharge and to sheep with poor care and nutrition without improving the conditions.
Consult your Veterinarian before use and if any unexpected effects occur. Keep out of reach of children.
PRECAUTIONS TO BE TAKEN BY THE PRACTITIONER AND WARNINGS FOR PHYSICIANS
All precautions should be taken against zoonotic diseases, and practicing veterinarians should use protective gloves during the application and removal of sponges.
HOW TO STORE AND SHELF LIFE
Stored in packaging, in the shade, in a dry place at 25°C. Shelf life is 36 months from the production date.
WARNINGS FOR END OF USE DISPOSAL AND NON-TARGET SPECIES
Sponges eaten by animals can cause disorders. Therefore, used sponges should be destroyed by burning. It should not be thrown into the environment haphazardly.
